Retatrutide vs Survodutide
Both retatrutide and survodutide are investigational next-generation obesity drugs. Neither is FDA approved. Retatrutide is a triple GLP-1/GIP/glucagon agonist; survodutide is a dual GLP-1/glucagon agonist. Retatrutide has Phase 3 topline data; survodutide has Phase 2 data. No head-to-head trial exists.
What Is Survodutide?
Survodutide (BI 456906) is a dual GLP-1/glucagon receptor agonist developed by Boehringer Ingelheim and Zealand Pharma. It activates two of the same three receptors as retatrutide (GLP-1 and glucagon) but does not include GIP receptor agonism. Phase 2 data showed approximately 18.7% weight loss at 46 weeks.
Mechanism Differences
Key Distinction: The Role of GIP
The key mechanistic difference between retatrutide and survodutide is GIP receptor agonism. Retatrutide activates GIP receptors (as does tirzepatide), which potentiates insulin secretion and may contribute to fat metabolism. Survodutide does not activate GIP receptors. Whether this difference translates to meaningful clinical differences in efficacy or safety is unknown without head-to-head data.
