Semaglutide Comparisons

Retatrutide vs. semaglutide (Ozempic/Wegovy) is one of the most common comparison discussions in communities. This page examines what the evidence actually supports — and what it does not.

Editorial note

No head-to-head trial comparing retatrutide and semaglutide exists. All comparisons are indirect and subject to significant methodological limitations.

Common Claims — Fact-Checked

These claims appear frequently in community discussions. Each is evaluated against the best available clinical evidence.

"Retatrutide is obviously better than Ozempic."

?Context Needed

Phase 3 data shows greater mean weight loss for retatrutide than semaglutide in their respective trials. However, these are different trials with different populations, protocols, and time points. No head-to-head comparison exists. 'Obviously better' overstates the certainty of indirect comparisons.

"Retatrutide has worse side effects than semaglutide."

?Insufficient Evidence

GI adverse event profiles are broadly similar across GLP-1 class agents. Retatrutide's glucagon receptor component may contribute additional effects. No head-to-head safety comparison exists.

Evidence Summary

Semaglutide (STEP 1): mean 14.9% weight loss at 68 weeks. Retatrutide TRIUMPH-1: mean 28.3% at 80 weeks. These are different trials — not a head-to-head comparison. Differences in population, dose, duration, and trial design limit direct comparison.

"Tried both — retatrutide is clearly superior to Mounjaro"

?Context Needed

Claim: Users who have used both tirzepatide (Mounjaro/Zepbound) and retatrutide report that retatrutide produces faster and greater weight loss, with some claiming it is "objectively better."

No head-to-head randomized controlled trial comparing retatrutide and tirzepatide has been published. The Phase 3 TRIUMPH-Obesity data (22.1% weight loss at 48 weeks) does exceed the SURMOUNT-1 tirzepatide data (20.9% at 72 weeks at the 15 mg dose), but these trials differ in duration, population, and protocol — making direct comparison unreliable. Individual self-reports are further confounded by dose differences, prior treatment history, and the absence of controlled conditions. The claim of superiority is plausible based on mechanism (triple vs. dual agonism) but is not established by clinical evidence.
